Dai et al.’s “Heat Shock Factor 1 Is a Powerful Multifaceted Modifier of Carcinogenesis” focuses on whether heat shock factor 1 (HSF1) contributes to the formation or suppression of tumorigenesis. In particular, environmental changes facilitate the adaptive physiological responses that help animals cope with certain stressors (Dai et al. 1006). Examples of proteotoxic stressors are acidosis, adenosine triphosphate (ATP) depletion, ischemia, and heat. HSF1s are the dominant stress-controlling cellular responses in mammalian cells.
Under controlled and stable laboratory conditions, HSF1 fosters longevity. For instance, the deficiency of HSF1 in a mouse suppresses skin carcinogenesis. However, when the mice are carrying p53R172H hot spot mutation, they produce a variety of tumor types, including carcinomas, lymphomas, and sarcomas (Dai et al. 1007).
