While you are encouraged to be polite to your supervisor of course, you should also be confident that you may know more about modern statistical ideas and methods than your supervisor (who is an excellent biologist, but hasn’t studied statistics for many many years, and who did ANOVAs with pen and paper when she last did). If you disagree with your supervisor, you should clearly and concisely explain to her why, and what you think would be better, and why it would be better. If you agree with her, then you should also explain why, and suggest any possible improvements. You may briefly suggest how experiments might be improved in future, but your main focus should be on the data and/or results you have already in each case and what can and cannot be done with them. Be as specific as possible.

The hypothesis of the supervisor is rejected. In a scenario where the humidity is low, the hatchings would be more female than male, and in cases where the humidity is high, the hatchings would be more male than female. 12 eggs were incubated in low humidity and the results were only 3 male crocodiles, this clearly shows that low humidity does not favor the male species. When the eggs were incubated at high humidity, we had 8 of the 11 eggs as male, the results indicate that when it is more humid the hatchings favor the male species as compared to low humidity. Statistically, in low humidity, male crocodile eggs account for a proportion of 0.25 while the number increases to 0.727 when humidity increases. The proportion test has a p-value of 0.0307 demonstrating a distinction between the egg sex ratio when crocodile eggs are incubated at varying levels of humidity.

I would inform the supervisor that One-way ANOVA with three levels of test will not be used to test if the proportion of males varies with temperature. One-way ANOVA is a statistical method for comparing differences in the mean between three or more independent variables (Ostertagova, Ostertag & Kováč, 2014). It can also be used to investigate the effect a single independent variable has on a dependent variable.
