Multiple Sclerosis (MS) is characterized by diffuse degenerative lesions in the central nervous system, specifically, in the gray and white mater. The focal damages in the meninges create toxic substances that cause direct or indirect demyelination via microglial activation, thereby causing neural and muscular disorders. This led to the discovery of following that can combat its signs and symptoms (Lassmann, 2018).
Spasmolytics
Spasticity is characterized by an increase in the muscular tone of the skeletal muscles, especially during high-velocity passive movement, thereby causing debility through the loss of the ability to do the activities of daily living (ADLs). The following drugs are used in the treatment of spasticity.
1 Baclofen
Baclofen, a gamma-aminobutyric acid B (GABAB) agonist, is a drug used to treat chronic spasticity. According to Katzung & Trevor (2015), it acts on the pre-and-postsynaptic membranes causing hyperpolarization of the membranes and, consequently, inhibiting the propagation of an action potential, thereby decreasing muscle contraction. In the presynaptic membrane, it acts by reducing the calcium influx and the release of glutamic acid, which is an excitatory neurotransmitter. By contrast, it works by aiding the inhibitory actions of GABA in the postsynaptic membrane (Katzung & Trevor, 2015).
