Hanging-Drop and Wet-Mount Preparations and Simple and Gram Stains

Coursework Instructions:

Answer questions on the sheet by creating a 2-3 page document.
Exercise 1: Hanging-drop and wet-mount preparations
1. How does true motility differ from Brownian movement?
2. What morphological structure is responsible for bacterial motility?
3. Why is a wet preparation discarded in disinfectant solution or biohazard container?
4. What is the value of a hanging-drop preparation?
5. What is the value of a wet-mount preparation?
Exercise 2: Simple stains
1. Define acidic and basic dyes. What is the purpose of each?
2. What is the purpose of fixing a slide that is to be stained?
3. Why are the specimens to be stained suspended in sterile saline or distilled water?
4. How does a stained preparation compare with a hanging drop for studying the morphology and motility of bacteria?
5. List at least three types of bacteria whose names reflect their shapes and arrangements, and state the meaning of each name.
Exercise 3: Gram stain
1. What is the function of the iodine solution in the Gram stain? If it were omitted, how would staining results be affected?
2. What is the purpose of the alcohol solution in the Gram stain?
3. What counterstain is used? Why is it necessary? Could colors other than red be used?
4. What is the advantage of the Gram stain over a simple stain such as methylene blue?
5. In what kind of clinical situation would a direct smear report from the laboratory be of urgent importance?

Exercise 1: Hanging-drop and wet-mount Preparations
In true mobility, there is a change in position regarding the original point. For instance, cells can flow autonomously and in a specified orientation. However, a Brownian movement is vibratory, which occurs at constant rates around a specific position.
In bacterial mobility, the morphological structure responsible for the movement is flagella. Possessing flagella on the exteriors of cells serves as a molecular drive or screw that generate thrust. Flagellum is a hairlike mechanism acting primarily as an organelle for cell movement.
Discarding wet preparation in biohazard containers is done to eliminate infections. That is necessary for laboratory safety as wet mount has living microorganisms. A small amount of bacteria suspension placed on the slide also help to produce clear specimen images under a viewing microscope.
Hanging drop is a complex method or procedure that allows long-term observations and a more vivid viewing of mobility. The technique is often carried out without the addition of stain. With a hanging drop, the organism cannot dry out rapidly and could live longer. On the other hand, a wet-mount preparation is valuable in helping to see living specimens and show the quality state of being motile, thereby helping in identifications.
Exercise 2: Simple Stains
A basic dye in microbiology is a positively charged stain inside a cell, while an acidic dye is negatively charged, affecting outside a cell. Bacterial surfaces repel acidic dyes, stain the background and leave a bacterial sell clear (Corrales et al., 2020). Basic dyes bind directly to stain negatively charged surfaces.
A slide that needs to be stained gets fixed to prevent washing off during staining. That can get done by passing a slide through a flame. Another alternative of fixation is flooding the slide with methanol, as this prevents liquid specimens from being washed off. The ...
