Introduction
Genome sequencing is a powerful technique to understand the genetic composition of prokaryotes and eukaryotes and how different species differ at both micro and macroscopic levels (Brown 2002). In genomics, different DNA mechanisms, internal structures, and different components are analyzed by extracting the genomes of the organism under study. Since such experiments are often complex, it is recommended to use DNA genomes from organisms that can easily be manipulated. In this laboratory experiment, the organism under investigation is Bacillus subtilis 1S53. This study aims at extracting the DNA gram-positive B. subtilis 1S53 since it is easier to avoid the chances of having spores that make it difficult to carry out DNA extraction experiments.
First, the cells will be opened up through different chemical treatments and DNA will be isolated from other cellular components, including organelles and other inclusions such as proteins. In most advanced experiments, DNA is always obtained as a pure macromolecule separate from RNA (Tan & Yiap, 2009).
