How Gene Regulation Occurs For Different Organisms
State that I generated the plasmids with motifs found in 5'UTR of rpl702 and hrd1 with cis regulatory motifs and did the experiments and find results as expected.
Thus, one aim of my experiment is to reveal distinct mechanisms of translational regulation, i. e. through cis regulatory elements, paired uORFs and potentially yet new mechanisms involving uORFs. With my undergrad peers, I generate luciferase reporter plasmids whose translation starts from 5' untranslated regions (UTR) of candidate genes as mentioned above. We transformed wild-type (WT) or gcn2-deleted yeast with these plasmids, grow transformants under amino acid starvation conditions and measure luciferase activity from the grown transformants. I am responsible for generating reporter plasmids carrying 5' UTR with uORFs. If Gcn2-depenendent translational control is confirmed with select 5'UTRs, I will generate reporter plasmids with the uORF start codon mutations and see if the uORFs identified are responsible for translational regulation.
