LAB REPORT ON EFFECT OF RESTRICTION ENZYME ON DNA PIECE
Introduction
DNA fragmentation is the separation of DNA strands into pieces. DNA is a hereditary material that carries genetic information in an individual responsible for development, growth, and reproduction and functioning. During DNA fingerprinting, the DNA strands have to be sliced on the target regions using restriction enzymes. Restriction endonucleases are enzymes that cut DNA strands at specific sites. The sites ate determined by a sequence of bases referred to as the palindromes. There are different restriction endonucleases that can be used and each enzyme will always have different cutting sites.
The restriction enzymes are classified into four type. They are different in their recognition sequence, subunit composition and cleavage composition. Type 1 cuts at sites remote from the recognition site. The type two cleaves within the recognition site or short specific distances from the recognition site. Type 3 cuts at short distances from the recognition sites while type 4 target the modified DNA. Some of the restriction endonucleases include: EcoRI, EcoRII, BamHI, HindIII, TaqI, NotI, HinFI, PvuII, and Smal among many others. There are also artificial restriction enzyme which can be produced by joining natural DNA binding domain to nucleus domain. These artificial endonucleases target large DNA sites and can also be designed to bind with the targeted DNA.
