The live, unstained cells of S. cerevisiae in the wet mount were far more visible under the conventional compound light microscope than the phase contrast microscope. It is because the compound light microscope has its own source of light in the base. In contrast, the specimen studied using the phase contrast microscope looked dark and transparent due to the brightness changes of the image. At the intracellular level, I found that the phospholipids, sterols, and sphingolipids in the plasma membrane were clearer under the conventional compound light microscope than the phase contrast microscope, giving me an idea that these components have a strong influence on the activity of proteins embedded or associated with the lipid bilayer.
