Glucose is a very important component in the nutrition because it is one of the three main ways the body gets calories or energy. Glucose is a simplest form of carbohydrates. Trinder assay is a method used to determine glucose concentration and identify given solutions. Trinder assay is a colour change test and is mainly used in medicine as a diagnostic test.
Beer-Lambert Law
This is the linear relationship between absorbance and concentration of an absorbing species e.g. glucose. The law is general written as
A=a (λ)*b*c where A is the measured absorbance, a (λ) is the wavelength dependent absorptivity, b is the path length and c is the analyte concentration.
Discussion
The law of Beer lambert is applied in the Trinder assay experiments since the concentration of glucose (which is an absorbing species) is tested in relation to its absorbance. In the process the enzyme GODPOD first reacts with glucose to form gluconic acid and H2O2.The reaction is initially very slow and this is because enzymes are very sensitive to both temperature and pressure and this enzyme had been stored in ice. The enzymes were stored in ice to prevent them from being denatured since they are very sensitive temperature and high temperatures can denature them.
