Abstract
The paper chromatography is the absorbent and filter paper where the different degrees of solubility in the solvents (methanol) allow separation when a solution thereof ascends by capillarity through a strip of chromatography paper in the solvent. The most soluble pigments will move faster and rise, while the less soluble pigments, advanced less in the paper strip. Bands of different colors indicating the pigments appeared, and from the top to bottom they were carotene, followed by chlorophyll a, then chlorophyll b, and lutein. Paper chromatography was useful in the separation of plant pigments and identifying these pigments.
Introduction
Chloroplasts have a mixture of pigments with different colors: chlorophyll-a (blue-green), chlorophyll-b (yellow-green), carotene (orange) and lutein (yellow) where lutein is a xanthophyll. Separation of photosynthetic pigments using paper chromatography is one of the common approaches (Barden-Gabbei & Moffitt, 2006). In the experiment, the aim is to extract these pigments, the products, as well as identify them. Methanol is the solvent and a comparison of the distance traveled by the various substances, and the solvent helps to determine the plant pigments.
Generally, the safety precautions are observed when handling methanol, and the green color of the leaves was contained after grinding in a test tube from where the pigment solution was collected. The strips of chromatography paper were cut and drops of pigment solution placed and spaced in time (Bettelheim & Landesberg, 2013). The test tube was added to the test tube and the chromatographies placed there to achieve separation. The pigment degradation was observed and recorded.
